No, Certificate of  Occupancy (U&O) is not required for a residential property sale. 

2. I have the property-owner signed contract, is that enough for a permit?

No. Willistown Township has a permit application for each specific job type. The property owner must also sign the Township issued application, and both the application and the signed contract must be submitted for review before a permit can be approved.
